Detection of protein dynamics using single molecule fluorescence confocal microscopy.

The ability to detect the fluorescence of a single fluorophore has led to great advances in understanding of heterogeneity and dynamics at the nanoscale level in life and material sciences. The specific emission properties of fluorophores like its intensity, decay time, emission wavelength (spectrum) and anisotropy can be used to probe changes in the fluorophores surrounding. For example, fluorescence decay time changes can be used as a powerful tool to monitor quenching and energy transfer processes. A confocal microscope setup will be developed with multiple avalanche photodiodes and a spectrometer (polychromator/CCD camera) that can be used to monitor changes in multiple parameters of the single fluorophore´s emission at the same time, providing a wealth of information. In order to extract as much information as possible, it is important to choose fluorophores with a high absorption coefficient, high fluorescence, low triplet, and low photobleaching quantum yield. For this, organic fluorophores (with and without the presence of photostability enhancing additives, like e.g. ROXS) will be tested and compared with a new class of silver cluster based emitters, attached to proteins in order to determine their photo stability performance. As a test case we will investigate the conformational changes that occur during binding and transport of substrate within the neurotransmitter transporter homologue, LeuT.

Qualifications
A MSc or equivalent degree in a field relevant to the research project and good English skills are required.

These scholarships are part of the Access to Masters (ATM) programme backed by the Convergence European Social Fund (ESF) through the Welsh Assembly Government.  Small and medium sized organisations located in the Convergence Area of Wales (Blaenau Gwent, Bridgend, Caerphilly, Carmarthenshire, Ceredigion, Conwy, Denbighshire, Gwynedd, Isle of Anglesey, Merthyr Tydfil, Neath Port Talbot, Pembrokeshire, Rhondda Cynon Taff, Swansea & Torfaen) are invited to participate in the muli-disciplinary initiative and benefit from a postgraduate research project tailored to meet the specific needs of the organisation as each MSc student will undertake a project assigned to an organisation. The School has a large number of fully-funded scholarships available for the following programmes: MSc Global Migration, Politics and Societ. 2011-Mazda Foundation Research Grants

The Foundation periodically identifies a priority area of focus. The Foundation’s current priority area of focus is Programs focused on identifying and/or remedying reading issues in children, with a particular interest in early intervention programs.Rather than inviting applications from a broad section of the public, the Foundation will each year identify potential candidates for grants in the Foundation’s nominated priority area. Each year the independent Board of Directors of the Foundation expects to identify a select number of projects for which an agreed contribution will be made to the approved organisation(s).

Although organisations should no longer apply to the Foundation for a grant, if an organisation has a project which relates to the Foundation’s current priority area, it may register a request to be considered by the Foundation by lodging a Project Notification in the form attached. Such notification may be lodged electronically or by hard copy to the Foundation.
